Objective To observe the expression of cytoplasmic polyadenylation element binding protein 4 (CPEB4) in glioma and the effect of CPEB4 on the growth of glioma U87 cells.
Methods The expression of CPEB4 protein in glioma tissues and normal brain tissues were detected.The expression of CPEB4 protein in cells, the cell proliferation, apoptosis, cell cycle and the expression of CPEB4, PIK3 CA, IGF2, SMAD3, IDH1 and AKT1 mRNA were measured.
Results The positive rate of CPEB4 protein in low-grade glioma gliomas tissues and high-grade gliomas tissues were higher than that in the normal brain tissues (
P < 0.05).The positive rate of CPEB4 protein in the high grade glioma tissues was higher than that in the low grade glioma tissues (
P < 0.05).The level of CPEB4 protein in the Low-grade glioma tissues and highgrade glioma tissues were significantly higher than those in the normal brain tissues.The level of CPEB4 protein in the high grade glioma was higher than that in the low grade glioma (
P < 0.05).At 24 h after transfection, the level of CPEB4 protein in the CPEB4-shRNA group was lower than that in the negative control group and blank control group (
P < 0.05).On the third and seventh day of culture, the A490 value of CPEB4-shRNA group were lower than those in the negative control group and the blank control group (
P < 0.05).The early apoptotic rate in the CPEB4-shRNA group was significantly higher than that in the negative control group and the control group (
P < 0.05).The G1 phase cells of CPEB4-shRNA group was significantly higher than that the negative control group and the control group (
P < 0.05), the S phase cells was significantly lower than those in the negative control group and the blank control group (
P < 0.05).The relative expression of CPEB4 mRNA, PIK3 CA mRNA, IGF2 mRNA and SMAD3 mRNA in CPEB4-shRNA group were lower than those in negative control group and blank control group (
P < 0.05).
Conclusion The CPEB4 is highly expressed in gliomatissues, CPEB4 involves the growth of glioma cells.The effect of CPEB4 on glioma cells may be related to the levels of CPEB4, PIK3 CA, IGF2 and SMAD3.
