Role of hydrogen sulfide in endotoxinemia and inflammation in rats with intestinal ischemic-reperfusion injury

  • Objective To investigate the influence of hydrogen sulfide on the levels of plasma C reactive protein (CRP),procalcitonin (PCT),endotoxin,TNF-α,IL-6 and IL-8 in rats with intestinal ischemic-reperfusion injury. Methods A total of 64 healthy male Wistar rats were randomly divided into sham operation group (A),ischemia-reperfusion group (B),ischemic-reperfusion and sodium hydrosulfide group (C),and ischemia-reperfusion and PPG group (D),n=16.The intestinal ischemic-reperfusion injury rat model was prepared as follow:used a midline incision for laparotomy,freed and clamped the superior mesenteric artery (SMF) for 60 min,and then reperfusion for 120 min.The tail vein injection of sodium hydrosulfide[100 μmol/kg]was performed 10 min prior to the onset of reperfusion,and continuous infusion[1 mg/(kg·h)]until reperfusion for 2 h in group C.The intraperitoneal injection of PPG (50 mg/kg) was performed in group D 10 min before the ischemia.The levels of CRP,PCT,endotoxin,H2S,TNF-α,IL-6 and 8 were tested;the synchronic blood culture was performed to evaluate the endotoxemia.H2S in the ileal homogenate was also measured. Results Compared with group A,the level of plasma endotoxin,PCT,CRP,TNF-α,IL-6 and 8 in the group B increased but H2S decreased.Compared with group B,the level of plasma endotoxin,PCT,CRP,TNF-α,IL-6 and 8 in group C decreased.Compared with C group,the level of plasma endotoxin,PCT,CRP,TNF-α,IL-6 and 8 in the group D increased.The level of plasma H2S in group D was significantly lower than that in group B and C.There were 14,6 and 15 blood samples respectively in group B,C and D infected by Escherichia colic.H2S negatively correlated with the levels of CRP,CT,endotoxin,TNF-α,IL-6 and IL-8. Conclusion H2S can decrease the level of inflammatory factor (PCT,CRP,TNF-α,IL-6 and IL-8) and attenuates endotoxinemia in rats with intestinal ischemic-reperfusion injury.
