Objective To investigate the effect of PARP-1 silencing on the cisplatin-resistance and related biological behavior of epithelial ovarian cancer cells.
Methods The A2780 human epithelial ovarian cancer cells was selected for the study and small RNA interference technology was used for silencing the expression of PARP-1.The Western blot test,RT-PCR experiments and MTT experiments were used to observe PARP-1 protein,mRNA expression and cell trends survival.
Results In the PARP1-siRNA transfection group,PARP-1mRNA expression was significantly lower than that of A2780 group and NC-siRNA transfected group(
P<0.05);PARP-1 protein expression in PARP1-siRNA transfection group was significantly lower than that of A2780 IOD group and NC-siRNA transfected group(
P<0.05);Compared with 0 μmol/L,the IOD of PARP-1 protein expression in 5 μmol/L and 10 μmol/L group were significantly lower(
P<0.05);With cisplatin concentration increasing,cell viability in PARP1-siRNA transfection group decreased fastest.
Conclusion PARP1-siRNA transfection can significantly reduce PARP-1 expression of human ovarian cancer cell line A2780,meanwhile the sensitivity of cells to cisplatin drugs and cell viability decrease significantly.
