Objective To study the impact of Tiaozhong decoction on motilin(MTL)and Stem Cell Factor(SCF) levels in the spleen deficiency with liver hyperactivity young rats with functional dyspepsia(FD),and discuss the possible mechanism of action.
Methods Total 36 clean grade SD young rats,age 21-28 d,weighing 50-70 g,were randomly divided into 6 groups:the normal control group,the model control group,the high dose group of Tiaozhong decoction,the middle dose group of TiaoZhong decoction,the low dose group of TiaoZhong decoction and the domperidone group,with 6 rats in each group.These young rats were fed regularly on the 1th week to adapt to the environment,then from the 2nd week,the rats(except for the normal control group) were fed irregularly,administrated with ice water(4℃) and received tail clamp stimulation to develop a rat model.The drugs were administered continuously for 14 days after the model was successfully established.The young rats in each group were fed nothing except water for 12 hours before the administration of medicine.Then,blood samples were collected from abdominal aortic and the he levels of SCF were tested by ELASE method.Afull thickness of gastric antrum wall was sampled to determine the MTL level by using immunohistochemistry method.
Results The SCF level in serum of the high and middle dose group were significantly higher than that in the model group(
P<0.01);The MLT level of the high dose group was significantly higher than that in the model group(
P<0.01),the levels between the middle dose group and the model group was statistically significant(
P<0.05).
Conclusion The model rat was successfully developed.Tiaozhong decoction can significantly improve the level of SCF in serum and MTL in gastric antrum.The up-regulation of MT and SCF may be the proper mechanism of Tiaozhong decoction in the treatment of spleen-deficiency with liver-hyperactive young rats with functional dyspepsia.
