Objective To explore the relative expression of lncRNA SOX2 OT in ovarian cancer cell lines and evaluate its effect on the biological behavior of ovarian cancer.
Methods Total 112 patients with ovarian cancer in Ningbo Yinzhou People's Hospital from April 2017 to April 2019 were selected as the research objects. The expression of lncRNA SOX2 OT in ovarian cancer, ovarian cancer adjacent tissues and ovarian cancer cell lines were analyzed. The effects of lncRNA SOX2 OT on the proliferation, cycle and migration status of ovarian cancer cells were tested.
Results The expression of SOX2 OT mRNA in ovarian cancer tissue(3.50±0.13) was higher than that in adjacent tissues(1.59±0.21,
t=81.045,
P<0.01). The expression of lncRNA SOX2 OT in HO-8910 PM cell line(5.78±0.04) was higher than that in HO-8910 cell line(1.05±0.11,
t=405.918,
P<0.01). The number of cloned cells, number of migrated cells, and number of invaded cells in the blank control group were higher than those in the siSOX2 OT-1 and siSOX2 OT-2 groups; the number of cloned cells, number of migrated cells and invasion cells in the negative control group were higher than those in the siSOX2 OT-1 and siSOX2 OT-2 groups(all
P<0.05). The expression of lncRNA SOX2 OT in endometrioid carcinoma, serous ovarian carcinoma, and mucinous ovarian carcinoma were 4.12±0.25, 3.68±0.42, 3.44±0.31, respectively, there was no statistically significant difference between the 3 groups(all
P>0.05).
Conclusion lncRNA SOX2 OT is abnormally high expressed in ovarian cancer cell lines. Knocking down lncRNA SOX2 OT expression can inhibit the proliferation, migration and invasion of ovarian cancer cells, and accelerate the apoptosis of ovarian cancer cells.
